I set my nginx.conf as the attached file

At first , I visit my site with no problem.
When I refresh my page for 4 times
It showed me 404 Pages not found, and I refresh it again but got the same message.

Is there something wrong in my config file ?

Need Help!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!
