Yes, very easily with 0.7.x and above. It would be something like
below ( check http://wiki.nginx.org/NginxHttpCoreModule#try_files )

location / {
try_files @varnish @application;
}

location @varnish {
// proxy to varnish
}

location @application {
// proxy to application
}
